the pathways responsible for the de novo establishment of H3K9me2/3 in C. elegans, a process crucial for preventing the harmful effects of unregulated transcription. Without proper regulation, the transcription of repetitive elements can lead to the accumulation of toxic R-loops and genomic instability. We aim to understand why transcription at some repetitive elements result in R-loop formation and if there is a feedback loop between R-loops/genomic instability and heterochromatin establishment.
