the role of FLASH/Casp8AP2 depletion in R-loop formation and its downstream effects on DNA damage. Initial findings showed that depletion of FLASH in mammalian cells induces g-H2AX foci, a marker of DNA breaks. The project aims to uncover the mechanisms by which FLASH depletion induces R-loops, investigate whether these R-loops activate DNA damage checkpoint signaling, and determine the importance of the R-loop-induced DNA damage response for S-phase accumulation. Further, the study will identify where in the genome R-loops form and explore the biological consequences of increased R-loops.
