“With the nucleus being packed full of DNA and RNA the potential for R-Loops to wreak havoc is enormous. The eruption of chaos is usually prevented by efficient mRNA export and degradation of aberrant transcripts. How this system works so well given the sheer number of different RNAs and steps in the process is fascinating to me.”

mRNAs need to be transported from the site of transcription, the nucleus, to the site of translation, the cytoplasm. Crucially only completely and correctly processed mRNAs should be exported, whereas aberrant mRNAs need to be degraded. This is ensured by maturity markers on the RNA which bind export adaptors which in turn lead to the recruitment of the central export regulator protein UAP56. For my PhD project I will biochemically, structurally and functionally characterize novel UAP56 interactors to decipher their influence on mRNA export or degradation.
